==== Adequacy of equipment and regulations ==== The factory was not well equipped to handle the gas created by the sudden addition of water to the MIC tank. The MIC tank alarms had not been working for four years and there was only one manual back-up system, compared to a four-stage system used in the United States. The flare tower and several vent gas scrubbers had been out of service for five months before the disaster. Only one gas scrubber was operating: it could not treat such a large amount of MIC with sodium hydroxide (caustic soda), which would have brought the concentration down to a safe level. The flare tower could only handle a quarter of the gas that leaked in 1984, and moreover it was out of order at the time of the accident. To reduce energy costs, the refrigeration system was idle. The MIC was kept at 20 degrees Celsius, not the 4.5 degrees advised by the manual. Even the steam boiler, intended to clean the pipes, was non-operational for unknown reasons. Slip-blind plates that would have prevented water from pipes being cleaned from leaking into the MIC tanks if the valves had been faulty were not installed and their installation had been omitted from the cleaning checklist. As MIC is water-soluble, deluge guns were in place to contain escaping gases from the stack. The water pressure was too weak for the guns to spray high enough to reach the gas which would have reduced the concentration of escaping gas significantly. In addition to it, carbon steel valves were used at the factory, even though they were known to corrode when exposed to acid.

== Pharmacokinetics == After inhalation of doses up to 800 μg (twice the maximum recommended dose) systemic blood levels of pirbuterol are below the limit of assay sensitivity (2–5 ng/ml). A mean of 51% of the dose is recovered in urine as pirbuterol plus its sulfate conjugate following administration by aerosol. Pirbuterol is not metabolized by catechol-O-methyltransferase. The plasma half-life measured after oral administration is about two hours.

==== Non-Hodgkin lymphoma ==== Results from several studies indicate that, compared to other autoimmune diseases, Sjögren's disease is associated with a notably high incidence of non-Hodgkin lymphoma, a cancer of white blood cells. About 5% of patients with Sjögren's develop some form of lymphoid malignancy. Patients with severe cases are much more likely to develop lymphomas than patients with mild or moderate cases. The most common lymphomas are salivary extranodal marginal zone B cell lymphomas (MALT lymphomas in the salivary glands) and diffuse large B-cell lymphoma. Lymphomagenesis in primary Sjögren's disease patients is considered a multistep process, with the first step being chronic stimulation of autoimmune B cells, especially B-cells that produce rheumatoid factor at sites targeted by the disease. This increases the frequency of oncogenic mutation, leading to any dysfunction at checkpoints of autoimmune B-cell activation to transform into malignancy. A study's findings concluded that continuous stimulation of autoimmune B cells leads to subtle germinal abnormalities in genes having specific consequences in B cells, which underlie the susceptibility to lymphoma.
Medications that have traditionally been known to lead to SJS, erythema multiforme, and toxic epidermal necrolysis include sulfonamide antibiotics, penicillin antibiotics, cefixime (antibiotic), barbiturates (sedatives), lamotrigine, phenytoin (e.g., Dilantin) (anticonvulsants), and trimethoprim. It was thought appropriate to choose a name to dissociate nicotinic acid from nicotine, to avoid the perception that vitamins or niacin-rich food contains nicotine, or that cigarettes contain vitamins. The resulting name niacin was derived from nicotinic acid + vitamin. Carpenter found in 1951, that niacin in corn is biologically unavailable, and can be released only in very alkaline lime water of pH 11. This explains why a Latin-American culture that used alkali-treated cornmeal to make tortilla was not at risk for niacin deficiency. The modern explanation is that alkali treatment enhances the bioavailability of tryptophan, not directly for any form of the vitamin. In 1955, Altschul and colleagues described large amounts of nicotinic acid as having a lipid-lowering property. As such, niacin is the oldest known lipid-lowering drug. Lovastatin, the first 'statin' drug, was first marketed in 1987.
== Procedure == Typically, the ammonium sulfate concentration is increased stepwise, and the precipitated protein is recovered at each stage. This is usually done by adding solid ammonium sulfate; however, calculating the amount of ammonium sulfate that should be added to add to a solution to achieve the desired concentration may be difficult because the addition of ammonium sulfate significantly increases the volume of the solution. The amount of ammonium sulfate that should be added to the solution can be determined from published nomograms or by using an online calculator. The direct addition of solid ammonium sulfate does change the pH of the solution, which can lead to loss of enzyme activity. In those cases, the addition of saturated ammonium sulfate in a suitable buffer is used as an alternative to adding solid ammonium sulfate. In either approach, the resulting protein precipitate can be dissolved individually in a standard buffer and assayed to determine the total protein content. The ammonium sulfate concentration added should be increased to a value that will precipitate most of the protein of interest whilst leaving the maximum amount of protein contaminants still in the solution. The precipitated protein of interest can subsequently be recovered by centrifugation and dissolved in standard buffer to prepare the sample for the next stage of purification. In the next stage of purification, all this added salt needs to be removed from the protein. One way to do so is using dialysis, but dialysis further dilutes the concentrated protein.
=== Energy and entropy === The microscopic features of liquids derive from an interplay between attractive intermolecular forces and entropic forces. The attractive forces tend to pull molecules close together, and along with short-range repulsive interactions, they are the dominant forces behind the regular structure of solids. The entropic forces are not "forces" in the mechanical sense; rather, they describe the tendency of a system to maximize its entropy at fixed energy (see microcanonical ensemble). Roughly speaking, entropic forces drive molecules apart from each other, maximizing the volume they occupy. == Theory == There are just 230 different ways of arranging objects in regular three-dimensional arrays. In molecular crystallography, these arrangements are called 'space groups'. However, only 65 of these arrangements are accessible to chiral objects or chiral molecules. The remaining 165 space groups contain either a center of symmetry or a mirror plane and are thus not accessible to natural globular proteins, which are chiral molecules. Wukowitz and Yeates developed a mathematical theory to explain the preference of globular proteins to crystallize in certain space groups. They suggested the preferred space group was determined by the number of degrees of freedom (D) or dimensionality as a measure of the ease with which a given symmetry can be formed. They analyzed the number of degrees of freedom for both chiral and achiral space groups where it was found that the space group P1(bar) with D=8 is theoretically the most dominant space group. Since the achiral space group had a higher degree of freedom compared to the chiral space groups, they predicted that racemic mixtures of protein enantiomers would crystallize more readily compared to the natural L-proteins alone by forming achiral {L-protein plus D-protein} pairs.
