Epitalon emerged from research conducted in Saint Petersburg by Vladimir Khavinson and colleagues, who studied short peptides as potential regulators of aging. The work built on epithalamin, a pineal gland extract reported to influence neuroendocrine function. Epitalon was designed as a synthetic counterpart with a defined sequence, allowing reproducible experiments that extracts could not support. Early publications described effects on melatonin rhythms and lifespan in animal models. These findings circulated mainly in Russian-language journals during the 1990s, which limited their visibility among English-speaking researchers.
The most widely cited claim is that epitalon activates telomerase and thereby extends telomere length. Supporting evidence comes largely from cultured human cells, where treatment was associated with increased telomerase activity and delayed replicative senescence. Telomerase activation is a biologically consequential effect, since the enzyme is largely silenced in most somatic cells. However, the route by which a short peptide would reach and act on the enzyme's regulatory machinery has not been established. == Treatment == Emerging therapies for genetic skeletal dysplasias include enzyme replacement therapy, small molecule therapy, hematopoietic stem cell transplantation and gene therapy. These therapies aim at preventing disease progression and thus improving quality of life. Enzyme replacement therapies are some of the mucopolysaccharidoses and Gaucher disease. Results have shown effectivity of enzyme replacement therapy. Hematopoietic stem cell transplantation can be lifesaving for some disorders, such as with malignant infantile osteopetrosis. Even with treatments such as enzyme replacement therapy and stem cell transplantation, people with skeletal dysplasia often require orthopedic surgery and other disease management interventions. There is a lack of information available to support these patients as most physicians may only see one or two skeletal dysplasia patients in their lifetime. Guidelines are available to support best practices for managing several areas of skeletal dysplasia, such as the craniofacial aspects of skeletal dysplasia, spinal disorders, diagnosis and management of type II collagen disorders, pregnancy of people with skeletal dysplasia, peri-operative management, and foramen magnum stenosis in achondroplasia. == Side effects == The safety profile of HMB in adult humans is based upon evidence from clinical trials in humans and animal studies. In humans, no adverse effects in young adults or older adults have been reported when HMB is taken in doses of 3 grams per day for up to a year. Studies on young adults taking 6 grams of HMB per day for up to 2 months have also reported no adverse effects. Studies with supplemental HMB on young, growing rats and livestock have reported no adverse effects based upon clinical chemistry or observable characteristics; for humans younger than 18, there is limited data on the safety of supplemental HMB. The human equivalent dose of HMB for the no-observed-adverse-effect level (NOAEL) that was identified in a rat model is approximately 0.4 g/kg of body weight per day. Two animal studies have examined the effects of HMB supplementation in pregnant pigs on the offspring and reported no adverse effects on the fetus. No clinical testing with supplemental HMB has been conducted on pregnant women, and pregnant and lactating women are advised not to take HMB by Metabolic Technologies, Inc., the company that grants licenses to include HMB in dietary supplements, due to a lack of safety studies.
The HER2 gene (also known as HER2/neu and ErbB2 gene) is amplified in 20–30% of early-stage breast cancers. Trastuzumab is a monoclonal antibody targeting HER2, inducing an immune-mediated response that causes internalization and recycling of HER2. It may also upregulate cell cycle inhibitors such as p21Waf1 and p27Kip1. The HER2 pathway promotes cell growth and division when it is functioning normally; however, when it is overexpressed, cell growth accelerates beyond its normal limits. In some types of cancer, the pathway is exploited to promote rapid cell growth and proliferation and hence tumor formation. The EGF pathway includes the receptors HER1 (EGFR), HER2, HER3, and HER4; the binding of ligands (e.g. EGF etc.) to HER receptors is required to activate the pathway. The pathway initiates the MAP kinase pathway as well as the PI3 kinase/AKT pathway, which in turn activates the NF-κB pathway. The HER (human epidermal growth factor receptor) protein, binds to human epidermal growth factor, and stimulates cell proliferation. In some cancers, notably certain types of breast cancer, HER2 is over-expressed and causes cancer cells to reproduce uncontrollably. HER2 is localized at the cell surface, and carries signals from outside the cell to the inside.
== In vitro activity == Like 'Peptide 3', NR58,3-14-3 is a broad-spectrum chemokine inhibitor. It however significantly more active than its earlier analogues migration inhibition potencies of 2.5 to 25 nM vs. CCL2, CCL3, CCL5 and CXCL12 with THP-1 monocytes, and vs. CXCL8 with neutrophils. In addition NR58,3-14-3 does not significantly inhibit leukocyte migration due to the other non-chemokine chemoattractants fMLP and C5a.

=== History === Black pepper has been known to Indian cooking since at least 2000 BCE. J. Innes Miller notes that pepper was grown in southern Thailand and in Malaysia starting in early 10th to the 11th century, when the South Indian kings began to extend their empire. The crop was brought to East Malaysia in 1840 by Chinese settlers; its most important source was India, particularly the Malabar Coast, in what is now the state of Kerala. The lost ancient port city of Muziris of the Chera Dynasty, famous for exporting black pepper and various other spices, is mentioned in a number of classical historical sources for its trade with the Roman Empire, Egypt, Mesopotamia, Levant, and Yemen. The ancient history of black pepper is often interlinked with (and confused with) that of long pepper, the dried fruit of closely related Piper longum. The Romans knew of both and often referred to either as just piper. In fact, the popularity of long pepper did not entirely decline until the discovery of the New World and of chilli peppers.
