Tofisopam (Emandaxin, Grandaxin, Sériel) is an anxiolytic that is marketed in several European countries. Chemically, it is a 2,3-benzodiazepine. While it may not be an anticonvulsant in and of itself, it has been shown to enhance the anticonvulsant action of classical 1,4-benzodiazepines (such as diazepam) and muscimol, but not sodium valproate, carbamazepine, phenobarbital, or phenytoin. Tofisopam is indicated for the treatment of anxiety and alcohol withdrawal, and is prescribed in a dosage of 50–300 mg per day divided into three doses. Peak plasma levels are attained two hours after an oral dose. Tofisopam is not reported as causing dependence to the same extent as other benzodiazepines, but is still recommended to be prescribed for a maximum of 12 weeks. Tofisopam is not approved for sale in the United States or Canada. However, Vela Pharmaceuticals of New Jersey is developing the D-enantiomer (dextofisopam) as a treatment for irritable bowel syndrome, with moderate efficacy demonstrated in clinical trials so far. Tofisopam is also claimed to be a PDE10A inhibitor, which may provide an alternative mechanism of action for its various therapeutic effects, and this action has been proposed to make tofisopam potentially useful as a treatment for schizophrenia.
=== Legal status === In February 2023, the Committee for Medicinal Products for Human Use of the European Medicines Agency adopted a positive opinion, recommending the granting of a marketing authorization for the medicinal product Hyftor, intended for the treatment of angiofibroma. The applicant for this medicinal product is Plusultra pharma GmbH. Hyftor was authorized for medical use in the European Union in May 2023. Sirolimus, as Rapamune solution, was approved for medical use in the United States in 1999; and as Rapamune tablets in August 2000. == Overdose == The median lethal dose of THC in humans is not fully known as there is conflicting evidence. A 1972 study gave up to 90 mg/kg of THC to dogs and monkeys without any lethal effects. Some rats died within 72 hours after a dose of up to 36 mg/kg. A 2014 case study based on the toxicology reports and relative testimony in two separate cases gave the median lethal dose in humans at 30 mg/kg (2.1 grams THC for a person who weighs 70 kg; 154 lb; 11 stone), observing cardiovascular death in the one otherwise healthy subject of the two cases studied. A different 1972 study gave the median lethal dose for intravenous THC in mice and rats at 30–40 mg/kg. A 2020 fact sheet published by the US Drug Enforcement Administration stated that "no deaths from overdose of marijuana have been reported."

Another author in 2008 raised concern that 2 randomized placebo controlled trials had been conducted, but their results remained unpublished for unknown reasons. None of the studies in the Cochrane review reported long term follow up after 3, 6 or 12 months post procedure. Another problem with some of the trials is that they were too small. The Cochrane review found that most trials reported short term improvement following the procedure, regardless of which material was used. In some studies in the control groups, even placebo (sham) injections and saline injections lead to patients reporting improvement. The authors concluded that due to the small amount of research available and its methodological weaknesses, further conclusions could not be made, especially regarding the long term effectiveness of the procedure. Another review which aimed to focus on the long term impact of the procedure included 889 patients across 23 studies. It reported a pooled improvement rate in measures of incontinence of 39.5%, on average 2 years after the procedure. In some cases there was no improvement after the procedure, and the injections needed to be repeated in up to 34% of cases.
